  1. Protein Intrinsic Fluorescence: Many proteins naturally possess intrinsic fluorescence, typically due to the presence of aromatic side chains like tryptophan and tyrosine. These amino acids can emit fluorescence when exposed to specific wavelengths of light.

  2. Thermal Denaturation: The sample is subject to a controlled temperature increase. With rising temperature, the protein undergoes thermal denaturation, causing structural and intrinsic fluorescence changes (and often changes in light scattering intensity).

  3. Fluorescence and Light Scattering Measurements: The instrument records changes in the protein's intrinsic fluorescence and light scattering intensity as it unfolds.

  4. Melting Temperature (Tm): The connection between changes in fluorescence intensity and temperature is used to determine the apparent melting temperature (Tm).
